Research Areas
TCR signaling and T cell development
The fate of developing T cells is determined by the strength of T cell receptor (TCR) signal they receive in the thymus. This process is finely regulated through tuning of positive and negative regulators in thymocytes. Recently, the Family with sequence similarity 49 member B (Fam49b) protein is discovered negative regulator of TCR signaling that has been shown to suppress Rac-1 activity in vitro in cultured T cell lines. However, the contribution of Fam49b to thymic development of T cells is unknown. In our lab, we are focusing on the effect of Fam49b on T cells developing, peripheral survival, and functional activity.
Obesity, inflammation, and the immune system
Chronic inflammation plays a critical role in the development of obesity-associated metabolic disorders such as insulin resistance. Obesity alters the microenvironment of metabolic tissues such as adipose tissue and liver from anti-inflammatory to pro-inflammatory, which promotes low grade systemic inflammation and insulin resistance in obese mice. These inflammatory changes are important because they represent potential therapeutic targets in obesity-induced metabolic disorder as well as obesity. In our lab, we are focusing on the mechanism of obesity-induced systemic inflammation caused by intestinal immune cells and development of novel anti-obesity treatments.

Research Areas
CRISPR-mediated Gene Therapy
The clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9 system has emerged as a powerful method for gene inactivation via the formation of small insertions/deletions (indels) and for specific genome editing by dramatically increasing the efficiency of homology-directed repair. The speed and ease of CRISPR/Cas9-mediated gene editing make it an ideal platform for optimizing in vivo disease correction. The ultimate goal of our laboratory is to develop new treatments for various human diseases through genome editing. Finally, we hope to provide the key to combating previously incurable diseases through gene therapy.
Stem cell-based Cell Therapy
Stem cell-based cell therapy is an important field of regenerative medicine with the ultimate goal of enhancing the repair machinery. Our study is based on induced pluripotent stem cells (iPSCs) and direct conversion providing convincing evidence that autologous cell replacement therapies can treat various disorders. To this end, our lab develops innovative cell therapy methods for clinical applications and possible solutions to incurable disease.

Research Areas
Our group at Chungbuk National University has been focused on the elucidation of structure-function relationship of protein-protein or protein-RNA complexes, especially nuclear transport related protein complexes including importin- family. A eukaryotic cell is divided into two major compartments, nucleus and cytoplasm by nuclear envelope. Intracellular signals are transmitted via the continuous and bi-directional transport of proteins between the nucleus and the cytoplasm through nuclear pores. To elucidate the molecular basis for such intracellular communication in atomic level, we crystallize and solve the structure of nuclear-transport carrier proteins.
Recently, we expanded our research interests with protein drugs including antibody. Collaboration with Osong KBIO for the X-ray structural analysis of virus antibody-antigen complex are in progress.

Total Synthesis of Natural Product
천연물 전합성(Total synthesis)은 간단한 출발 물질(Starting compound)로부터 유기화학 방법론을 활용하여 천연물을 합성하는 기술이다. 천연물로부터 신약을 개발하기 위해서는 분자구조와 생리활성 관계(Structure-Activity Relationship)를 검색하는 과정과 최종 목표화합물을 저비용으로 대량 합성할 수 있는 효율적인 과정의 개발이 필요하다. 이러한 과정에서 천연물의 효율적 전합성 기술과 응용이 필수적인 역할을 하게 된다. 실험실에서 이루어진 최근 연구결과, 다양한 구조를 가지는 천연물 전합성을 완료하였다. 대표적인 예로서는 Chloropeptin I, Fluvirucinine A1, (+)-Brefeldin A, (S)-Rodgersinol, Obovatol 등이 있다. 이들 천연물들은 독특한 구조적 특징 및 흥미로운 생리활성을 가지고 있으며, 전합성을 위하여 새로운 합성전략의 수립과 도출된 문제의 해결 등을 통하여 성공적으로 전합성을 완료하였다. 현재 Methyl Honokiol, Tetrabenazine 등의 천연물을 전합성 중에 있다. 천연물 전합성 능력/기술은 다양한 구조의 천연물 유도체의 합성에 활용될 수 있다.
Medicinal Chemistry & Drug Discovery
세계적 신약개발 현황은 치료질병의 표적분자의 구조 및 약리작용단 (pharmacophore)을 기초로 한 합리적 약물설계 (rational drug design)에 의해 주로 진행 되고 있다. 합리적 약물설계를 위해서는 표적분자의 구조분석, 컴퓨터 분자모델링, 약리작용단의 분석 등의 의약화학적 신기술이 요구된다. 이러한 합리적 약물설계 기술을 이용하여 최근 항암제, 혈과평활근 증식 저해제, 치매치료제 등을 포함한 다수의 신약 선도물질을 개발하고 있다.
